On December 2rd, a nasal spray COVID-19 vaccine, VectorFlu ONE, was approved for emergency use by the national authorities in the Chinese mainland. The vaccine has been jointly developed by Hong Kong University, Xiamen University and Wantai Biopharmaceutical Company. It is reported to be the first Covid-19 vaccine in the world in the phase of clinical trials with proven safety and broad-spectrum efficacy in phase III clinical trials to date. According to the interim primary data from South Africa, the Philippines, Colombia and Vietnam, the vaccine provided good protection against Omicron variant strains BA.2, BA.4 and BA.5. The data demonstrated that the vaccine provided good protection against Omicron variant strains in both the baseline immunization for people without immunization history and the sequential booster immunization for people with immunization history.
The nasal spray vaccine was developed by inserting a RBD fragment of Spiking Protein of Coronaviruses into the vector of double attenuated Influenza A Virus, which has been specially modified to improve safety and efficacy. Bearing a striking resemblance to Influenza Virus, Covid-19 (especially the Omicron variant) is featured an anatomical distribution of susceptible cells throughout the respiratory tract from the nasal cavity. Aimed at this feature, this vaccine can form a first-line immune barrier against invasion into the respiratory tract by intranasal spraying, which mimics the natural infection of viruses. It complements the systemic protection mechanism induced by intramuscular Covid-19 vaccine for a more comprehensive protection.
As the study reveals, the nasal spray has been proved to prompt a multidimensional protective immune response including cellular, humoral, innate and trained immunity. Thus it provides broad-spectrum protection, largely unaffected by virus antibody escape mutations. It prompts the equivalent protective immune response to the prototype strain or the major variants, including the Omicron BF.7, XBB and BQ.1.1 variants.
